The last row of Fig. SM2 shows muscle-contraction induced changes in the intensity of staining of LC spots. In resting muscle the last peak, the major isoform phosphorylated contains as much as 63% of the total intensity, which decreases to 35% in hiring, muscle. At the same time, the intensity of 23% in the previous peak of resting muscle increases to 49% shrinkage. This is an example of a simple method (but not sensitive) to track changes in the phosphorylation of LC during contraction without using 32P-labeled muscle.
